If you restore from backup (from MAC), will it erase photo's / will they be lost - and emails before they're backed themselves`

If you restore a backup for an iPhone from an iMAC, will it erase photo's that have not been backed up, onto the iMAC

Yes. Restoring a backup overwrites everything on the phone. If you have synced (not backed up) data to iCloud, such as photos, you can resync them to the phone after restoring the backup.

Note that anything synced to iCloud (Settings/[yourname/]/iCloud) will not be included in backups. So to deal with your situation turn on iCloud sync for photos, wait for sync to complete, then restore the backup.
