It doesn’t matter. Lithium ion batteries are not picky about the first charge. You should try to leave it plugged in while first setting it up, because the setup process can use a lot of energy, but there is no need to charge it before setup if it is over 50%.
Is it good to leave the phone to charge to 100% at night or to charge to 80% before going to bed and not to charge all night

I'd say just do whatever works well for you. But if you leave it connected to power, it will eventually charge to 100% (at least on the display) by morning at the latest unless there are active steps taken to disconnect before it gets there. I don't see any reason not to allow it to charge up to 100%. It's just one of those things where it gets really tedious to follow some "rule" that may result in somewhat better longevity at a cost of utility. Apple's "optimized charging" is attempt to marginally improve battery longevity in a way that won't affect how most people use their devices. It's supposed to learn how it's used. If someone wakes up every morning at 7 AM and removes an iPhone from power, it's supposed to learn the pattern and then charge up to 100% starting maybe 30 minutes before. But if the pattern is broken like (something I've personally done) waking up at 3 AM for an early morning flight then it's not going to know that and will be at that 80%.
That being said, I put my phone in my pocket, use it when I need it, and charge it when it's convenient. I have optimized charging enabled, but I don't try to follow a pattern to force it to do its thing. There's a point where it's just taking steps too far given that it's relatively inexpensive to get the battery replaced.
