Yes, dust and debris can enter the earpiece as well as speaker, mic, and charging ports at the bottom. Apple Technicians use something called "Blue-Tack" which is just tacky blue putty to clean out the ports. You can find this at Walmart.
If you're near an Apple store they do this for free. But if you're having Audio related issues, I'd consider making an appointment at the Genius Bar so they can run an Audio test.
