With update to 12.12.6.1, I no longer see option to backup my iPhone
How can I backup my iPhone to PC using iTunes 12.12.6.1? I see no options to backup now

Is your iPhone "visible" to your iTunes 12.12.6.1?
Should be this case, click on the iPhone icon and you'll see the "backup now" option in the same place it was before (unless your iTunes update was unsuccessful).
