I've shared my contact card using my iphone 12 with numerous business partners & I've recently changed my number. If I update my contact card to include my new number, will it automatically update it for all of those that have saved my contact card ?

Your contact information would not update automatically on other people's devices. You would need to share your updated contact card again using these steps from Add and use contact information on iPhone:
Share a contact
Tap a contact, tap Share Contact, then choose a method for sending the contact information.
Sharing the contact sends all of the info from the contact’s card.
