Are you referring to displaying the Quick Notes note when you swipe from one of the corners of your iPad Pro’s screen? IF YES, then:

Open Settings ➡️ Notes ➡️ Corner Gestures ➡️ Allow Finger to Swipe from Corner: Set this switch to ON and select Quick Note for either the right or left corner.

Do you have an iPad model that supports iPadOS16’s Stage Manager feature - and if so, is Stage Manager enabled?

If Stage Manager is enabled, the QuickNote corner gesture is not available - and will be “greyed-out” in iPad settings. Stage Manager and QuickNote features are mutually exclusive - and cannot be used together.

To use QuickNote in iPadOS16, Stage Manager must be disabled:

Settings > Home Screen & Multitasking > Stage Manager > Use Stage Manager on iPad - set to OFF
