Is the initial quick start a one time thing or is there a way to do it again if the data transfer from the existing phone failed?

Transferring from iPhone 6s to iPhone SE third gen

Yes. You can perform Quick Start again. It is wise to perform, if it is was not 100% successful the first time around.
