Had exact same error and followed your advise by deleting Genius file and it worked perfectly. One more iTunes error resolved and several more to go. Thank you very much for the helpful advise!

This seems to be an ongoing issue that need a permenant soltuion from Apple. The solution of deleting the genius file works but in my case, this needs to be done each time iTunes is restarted. What gets me is that there was an update earlier this year that seemed to fix this issue but then another update came out within a week or so and now I'm back to deleting the file everytime I want or have to restart iTunes.
Try the following:
1. Go to the following folder: username/Music/iTunes/
2. Remove the following files from this folder:
iTunes Library Genius.itdb
iTunes Library.itl
iTunes Library Extras.itdb
3. Open iTunes.
4. You will have to re-scan your media.

No! Not the iTunes Library.itl file!! That is what makes your library. Delete that and when you open iTunes you will get a blank, brand new library which will certainly take care of the issue because you will be starting from scratch (the media files will still be on the computer). By "re-scan your media" this will mean adding it all again to the library, rebuilding your playlists, starting off with playcounts zero and no ratings.
Caddyfan, I see you have systematically posted in quite a number of topics about this issue. I hope you post a correction about this in each one, or if you did indeed mean delete the entire library file at least make it clear this will be a consequence of your suggestion.

Just offering this suggestion.
Unfortunately in my case, iTunes would not launch on my iMac until all 3 of these files were removed.
Perhaps this is a last resort option for some cases.
Deleting Genius file works temporarily but the fault returns.
It also won't let you delete the genius file if iTunes is open. This causes a problem if you have just ripped/added tunes to your library because they won't be saved and you have to shut down itunes to delete the file.
keeps recreating the said files every time iTunes is opened, even though Genius is disabled.

Be very careful if you delete iTunes Library.itl I had to spend weeks trying to get my library back I lost all play list ratings and how many times a song had been played. I never was able to get the information back even with 3rd party apps from my ipad, Iphone, & Ipods.
