My son and I have been using Apple One Premier for the last few months and I want to add my husband to our family subscription but he is afraid he will lose all his Apple Music playlists and preferences. Will adding him affect his Apple profile/preferences?

No. Adding him will allow him to use the Apple Music subscription from the Apple One bundle, but will not affect anything he currently has.
Do not cancel his own Apple Music subscription. Just add his Apple ID to your Family Group, and it should seamlessly transition, and retain everything. It will automatically cancel his subscription once he joints the Family Group.
