My iPhone 14 Plus has a dual sim: 1 for business, other for personal.
Would like to set up an auto reply to clients on the business line specifically for days I’m “closed”
Running 16.6.1 iOS

Currently, that feature is not available on the iPhone. In iOS 17, you can assign different ringtones for each active line ,however, aside from that there is not much that you can set that isolates one line from another.
There is one other option and that is that you may be able to setup something from your carrier whereby you can push all incoming calls directly to voicemail on that line during non-business hours. Check with your carrier whether they have such a feature.
