I often use the color filters accessibility setting for red tint at full intensity in dark environments, but it is sometimes hard to see contrasts I’d expect. For example: red and white are indistinguishable. Is there a way to preserve value/shades like a monochrome b&w filter but with a red overlay instead of white?

As you have learned, there are preset filters with no manual adjustments for base color and overlays.
Have you tried using the Night Shift feature in Settings under Display & Brightness?

I am not aware of any but that is just because I do not use that feature. I recommend that you perform a search on the internet and check whether such third-party apps exist for your requirements.
