Generally speaking, no.
According to Apple, your Apple Watch must be connected to WiFi for it to receive software updates:

You can try connecting your Apple Watch to your iPhone's hotspot, but I cannot guarantee that this works. According to Reddit and previous Apple Support posts, the update attempt usually fails.
