why can't I press percent sign , even by pressing Shift & 5 together ?

Does shift work with all the other keys?
Are you talking about pressing shift and 5 on your Macbook air keyboard? An iPhone keyboard doesn't have that kind of layout.

I don't think there is any setting that would cancel normal output from shift 4 and shift 5.
