is it possible to back up to icloud with a broken screen and not able to unlock the phone? My son's phone has totally lost the screen and it's not done an auto-backup since 2023, is there a way of backing up via iTunes from my laptop?

Sorry, but no, it would not be possible to back up in any way.....iCloud, computer, laptop etc. with a broken screen. The same would be true if the only problem was that the phone was locked.
Once the phone is repaired and reset, you could restore using the 2023 backup, but then the phone would look like it did at the time.
Whether the cost of repairs (the phone will probably also need a new battery) would be better spent on a new phone will be your call.
