What do you mean with "reset"? I don't know of any way to get it back to the default state since there isn't much in the way of user control.
try searching in the forums for the answer. I am pretty sure someone gave the Terminal commands for it.
Well how do I sort by letter? Because I have apps out of order...
Yes....but will that make it stop re-arranging all my icons every time I wake it up or restart?
Until that gets fixed, LaunchPad is a waste of hard drive real estate.

The Terminal hacks above are the main cause for Launchpad re-arranging your icons.
The hack above corrupts the database as it deletes records without paying any attention to even basic SQLLite concepts (triggers, foreign keys, etc)
Alas, while this may be true....I have not tried any of the Terminal hacks above. Here we are how many weeks later and this problem has yet to be addressed and yet continues to make LaunchPad the greatest Finder utility there never was.

1. To reset the Launchpad open Terminal: /Applications/Utilities/Terminal
2. Open Terminal and simply type: killall Dock, press the retun key.
3. Open Launchpad and notice all of your Applications are back in default alphabetical order.
* Executing the command that is listed above is the proper and most accurate way to reset Launchpad. Upon executing this command it will NOT cause any system corruption.

Nope, that doesn't work, in Mountain Lion at least. It killed the dock, but when it came back it still had my apps in my original sort order.
This is a perfect solution (XLine app) for a lot more than only launchpad troubles. Thanks a million
Unable to find it (server issues) is the app somewhere else?
Install free version of Launchpad program, http://launchpadcleaner.com, run it, and click restore all items to default.
Also this will help if you have a problem when icons in Launchpad disappears, or you want to delete some of them.
I try it and it works.
