Now all I get is a spinning beach ball and iTunes locks up.

Same problem here, but with an iPhone4. iPad2 has no problems

I thought the issue might be with my Snow Leopard Mac Pro. So I tried on my MacBook Pro Lion and get exactly the same problem.
Complete freeze with a beachball. Only ways to unfreeze are to force quit iTunes or disconnect the iPod Touch.
Same thing here with iPhone 4: connect and spinning beach ball. I can't sync at all.
Let me add the same problem. iTunes 10.5 works fine until I plug my iPhone 3 into it, then I get an endless beach ball and nothing in iTunes functions.
Running a 2011 27" I7 iMac, OS X 10.7.1
it doesnt work with iPhone 4 on Lion (macbook pro) either.
This is ridiculous. How can a bug this big go unnoticed?
Apple better fix this ASAP, I'm sure there will be plenty of people trying to sync their iPhone 4 before getting the 4S.
Inexcusable.
New iMac (actually my first) running 10.7.1. No problems installing iTunes update or syncing with an iPhone 4 or an iPad (both with latest firmwares). So perhaps this is not an "inexcusable glitch", but something that some users are having a problem with. Don't see how it is possible to find every "glitch" in software - even with Apple's more controlled system - as opposed to Microsoft where you can install whatever hardware/software you wish.
I left it on the spinning beachball for about 30 mins and it eventally synced.
Same problem here with 10.6.8 and iPhone 4 on 4.3.5.
iTunes just keeps getting slower and slower, and more unstable. It's beginning to need a massive overhaul.

Thanks for this. I let mine sit and eventually it became responsive again and allowed me to sync.

after letting it sit for about 10min it unfroze and synched the iphone
after disconnecting and connecting again it froze for about 15sec and then synced.
Looks like a similar issue a few years back where the sync froze too.
would still need a bugfix, even it only effects some machines.
Others connect and let it run unattended so they might not notice.
Same problem. Going to leave it connected to see what happens.
My iphone 3g and ipad 2 are not being recognized at all by itunes 10.5... (Windows 7)
iTunes really is Apple's red herring...
Same problem, left it connected and it unfroze, now no problems.
Side note, my phone rang as right after I connected my phone to itunes the first time (before i noticed the freeze)... maybe just a coincidence.
