It won't harm your iPad to leave it in sleep mode. The battery can drain, particularly if you have an app open that communicates (to avoid that, close them fully from the task bar by double tapping your home button and the task bar will open, then hold your finger on any of the task bar apps until they all jiggle and touch the ➖ on the upper left side).

I personally, shut my iPad completely down when it's not going to be used for several hours. But, the choice is yours.

Either. Like some have said, if you have an app that is running - like if you have a regular 'push' for your e-mail you'll possibly get a little drain overnight. Or if you have an app that's running that has a sound (say your mail chirps everytime you get a new one) then it can be an issue.

However I've done it both ways. Most of the time, I just let it sleep. Sometimes I'll turn it off. So a personal preference thing. I think the only time you'll have a major issue is if you set your ipad so it won't go to sleep and your batttery drains totally.

now I am of the mindset that every comptuer needs to be turned off and rebooted every so often. just to clear out the 'cobwebs' so to speak. But it's not mandatory. I've gone days and days just letting my ipad sleep, same with my Touch.
