Ok so I know this is weird, but mine started working again. At the start, I tried everything. I tried a hard reset, I ended up restoring my phone from an old backup, I even restored it and tried from scratch, I tried as much as I could, but it wouldn't work.

I went into the shop, my provider said that I have to take it up with Apple. I ring apple, I organise for the issue to be looked at. I was busy that day, and then my phone ran out of battery itself. (which never really happens)

When I charged it and turned it on, it was fixed!

Since then, it works the majority of the time. I never ended up pursuing it with the Apple store any further than calling them at the start. Sometimes it doesn't work, but I just go into YouTube and its working again. Its really weird.

Can you guys please try this as well so we can see if it works for everyone?

I have also lost sound on my iPhone 4S running iOS 5.0.1. I always lose sound whenever I place the phone in silent mode using the switch on the side - when I click the switch off the sound is gone. I also frequently lose sound when, instead of placing the phone in silent mode, I turn the volume all the way down. When I try to turn the volume back up there is no sound. At other times the sound just stops working. So far, whenever I have lost sound forcing the phone to reboot always restores the sound.

This is what I have found. When I do my long runs and use headphones, the sweat travels down the wire and into the headphone jack. When the headphones are removed, the phone still thinks the headphones are plugged in causing no sound to come out of the speakers. Get a small piece of tissue paper (1.5" x 1.5"), twist one corner of it so that it is roughly the same diameter as the headphone jack and about 1" long. Insert it into the headphone jack while your spinning it in the direction of the twist and voila, phone is fixed. Make sure you turn the phone off when you do it.
