Version 10.6.1 no longer allows use of the space bar to start and stop the currently playing music. Also, the return key can no longer be used to strart a song. I find it very convenient to use the space bar to stop and start the music.
MacBook Pro (13-inch Mid 2009), Mac OS X (10.6.8)

I can only replicate the problem if I click in the Search field.
If I click elsewhere, the Space bar works correctly to start/stop a song.
If it's not working for you, I suggest you contact Apple.
I still have the same problem, pressing the Space Bar to Start and Stop iTunes does not work anymore. I have an Intel Mac mini running OS X 10.5.8. Somewhere along the way iTunes updated to 10.6.1 and I noticed that I could no longer use the Space Bar to Start and Stop iTunes. I removed the iTunes 10.6.1 app from the Applications folder, and downloaded the latest version of iTunes 10.6.3 and installed it. When I opened iTunes 10.6.3 it still loaded my old prefences file and recognized my music library and personal settings, but i still cannot use the the Space Bar to Start and Stop iTunes. I also am having similar problems: • Cannot Sign In to iTunes from the Store menu, • cannot Authorize this computer, • cannot Deauthorize this computer in iTunes, and I cannot even • Check for Availble Downloads in iTunes. I did Deauthorize this computer prior to removing the iTunes 10.6.1 app. I am beginning to suspect that iTunes 10.6.x is not fully compatible with OS X 10.5.8.

What has fixed the problem for me is using the ..."you can enable keyboard commands for all controls by pressing Control-F7 or selecting the "All Controls" radio button in the Keyboard system preferences.".
Pressing Control-F7 does in fact toggle the keyboard shortcuts off and on.
However, I am NOT able to find the "All Controls" radio button in the Keyboard system preferences.
