I do lots of presentations on my MacBook Pro. I would like to turn off notifications when I present, but still keep all my notification preferences. Is there a way to toggle notifications on and off?

You can 'option'-click (hold down the option key when you click) the Notifications icon at the top right of your screen to toggle on and off.
If you connect your Mac to an external monitor, it will do this for you automatically.
Yes, Go into the Notification area by clicking the icon on the menubar far right.
Scroll down, you'll see a snooze for notifications that will turn them back on tomorrow or until you turn them back on.
Opt-Click on the notifications icon in the menu bar will set the notification center inactive for the rest of the day. The icon will be greyed out for that time.
If you click the Notifications icon in the menu bar and then scroll up a little you'll see a switch that allows you to shut down Notifications for 24 hours.

Is there a way to permanently turn them off? I don't want the 24 hour delay...I want them off permanently.
I've already tried removing them from the notification center, but even then I still get Mail alters and calendar alerts.

Have you tried selecting none as the alert style in System Preferences?
Try going to desktop and screen saver and removing it from corner activations, I had to turn mine off to play some DOTA.
Onyx has an option to disable notifications for good, as well as several other tweaks (which probably can be done through a terminal command).
It's great!
