Open System Preferences > Security & Privacy then select the General tab.

Click the lock icon bottom left corner of the pane to make changes. You may be prompted for your admin password.

Now select: Mac App Store and identified developers

Click the lock to prevent further changes and try the App Store.

You may also need to disable anti virus software and turn off the Firewall in System Preferences > Security & Privacy > Firewall

I've been having problems connecting to the App Store (iTunes Store worked though) with a brand new MacBook Pro. The solution was to repair permissions on the hard drive. To do this, open Disk Utility (under Applications\Utilities), click on the hard disk, then on “Repair Disk Permissions”. It will take a couple of minutes to complete and might resolve the problem. Good luck!
