Try adding the DNS Servers: 8.8.8.8 & 8.8.4.4
Open System Preferences > Network > (Select your means of connecting to the network) > Advanced > DNS
Click the +
8.8.8.8
8.8.4.4
Changing DNS servers will not help. I'm having the same problem, and my logs don't show any indication of DNS lookup errors. My logs show a 503 error, which is a Service Unavailable error. See:
http://www.w3.org/Protocols/rfc2616/rfc2616-sec10.html
The only fix likely to work is to simply wait for a day or two until the issue is resolved, and try again.
Same problem NSURLErrorDomain-1100. I have a suspicion that the updates related to the App store are flawed. Given the changes, updates and the amount of code generated , human error may happen. Patience is a good idea.

I've had the same error (NSURLErrorDomain error -1100) consistently since yesterday evening (living in Sweden) and this afternoon it still won't connect to the update servers correctly.
However, when connecting through my VPN service (which gives me an IP in the UK) I can connect just fine and update my software through the Mac App Store without problems. Definitely a problem with some of Apple's regional update servers.
Same problem here in Italy (osx 10.8.1).
Same here in Panama. Same error since yesterday afternoon
if you want to update to the new iTunes, you can do that from here:
http://www.apple.com/itunes/download/
saludos desde El Salvador !!!
Lo mismo por aquí, por Canarias.
Same for Greece, I tried a zillion times after the event finale and no luck.
Like other people said, there are two (three) options from this problem,
-Download iTunes from here: http://www.apple.com/itunes/download/
-Use a VPN to connect an other Servers like UK, then download.
-Just wait two or three days.
Using the DNS from Google, don't solve the problem. In fact, the problem resides in some US Servers. But if you change your IP (with VPN service), use another country, like UK per example 😉
