how do i use an iTunes library in a disk other than startup?

I boot off a Solid State Drive (only 240 GB) but have most of my data in a 750 GB HDD. My media has grown to the point that I cannot fit it into the SSD but easily fits into the HDD. Can I just use the iTunes library in the HDD?
