Is it possible to connect a non-Apple laptop (Lenovo) which has a mini display port to my iMac 21,5 inch (model late 2012). I want to use the bigger screen of the iMac.

No, the Lenovo would need a Thunderbolt connection that has the ability to pass through video, it's doesn't have that. Please read Apple's article on Target Display Mode, please click Target Display Mode: Frequently Asked Questions (FAQ)
Thanks for your comment. And unfortunately the Lenovo doesn't have a thunderbolt connection.
If you had a 2009 or 2010 27" iMac with Mini Display port it would work, however in 2011 Apple went to the much more capable Thunderbolt technology.
