Is it safe to do software update without backing up my iPhone first? I am at wok and think I should do my software update, but don't have my laptop computer with me to backup my iPhone. My battery seems to be draining slowly without even using it. Any suggestions would be greatly appreciated.

If you don't have a backup, then no. It is not "safe". It is possible to set up iCloud backups as described above, but you should check to make sure it's on and working if you intend to rely on it, or check to make sure it's been backed up in iTunes on your computer.
You should NEVER attempt an OS update without ensuring you have a good, current backup. That applies to any platform, not just iPhones.
You would have to select to backup to iCloud. It does not automatically do it, unless you have set it up to do that.
You should ALWAYS backup before any update.

Backs up to icloud you can do almost everything over your wifi
I haven,t used my pc for iphone/ ipad for over 2 years.
So yes do softwear update over your wifi.
