As far as I know you could only do that if you can find a region that uses those days as the weekend and change the region settings in Settings>General>Language and Region. This would change a whole bunch of other settings as well, though.
Well I tried that since it worked so well on my MacBook Pro but nothing changed. I turned the calendar off, changed the region, and turned the calendar back on. Sat/Sun were still highlighted. Very frustrating.
Search around in the App Store for other calendar apps. Probably start with an internet search for something like "iphone calendar app flexible weekends" and see what pops. As all the calendar apps draw from the same database, you don't have to enter your info all over again.

The app "Week Calendar" in the app store has a feature to select any days as "weekend". Plus a lot of other useful features.

I haven't found much missing that I currently use. But comparing it to Pimlical (http://pimlicosoftware.com/),which is available for Windows, Mac and Android, but not iPhone, there's a lot that's missing. Pimlical implements its own reminders, and allows hundreds of customizations. And all of the revenue from sales goes to support Gorilla conservation.
The features of Week Calendar that I use daily include event templates, drag and drop rescheduling, multiple alerts per event, event icons (which iCal on a Mac recognizes, but does not support), Multiple views, extended search (as discussed)...
You can change which day of the week the calendar begins on in Settings | Mal, Contacts, Calendars but Saturday and Sunday will always be highlighted regardless of which day you begin the week on.

I used to use the templates quite a bit.
I'll have to give Pimlical a try. Just got a new Samsung S6 Edge+ from work.
