Will I lose my iPhone 6 iTunes backup on my Mac if I update to new version of iTunes? Can't set up my new iPhone because told I need to download iTunes 12.2.2.

No. Of course not. If that happened, then it would happen to millions of iphone users all over the world each time an update is released, and do you really think Apple, or any other company would design software that would cause that to happen?
