I had problem with this on my 6s Plus too with iOS 10.0.2.
However, upon reading here (http://osxdaily.com/2016/09/26/take-screenshot-ios-10/) it works well with me now.

I found on my iPad Air 2 that both Home and On/Off must be pushed quickly SIMULTANEOUSLY! The old way was more forgiving. I made it work twice so far.

Here's the thing: we're talking about a function I use regularly. Then after a major update, SUDDENLY functionality was only sporadically successful. Obviously I'm not alone; many other users have reported a similar problem. The fact it hasn't been experienced by EVERY user is certainly not an indication that it is a shared hallucination. You can keep trying to insult people who are having the problem — who knows? maybe that will culminate in a solution somehow.
The Assistive Touch alternate method is a temporary remedy that works when the primary method fails. This is helpful.
My device is completely up-to-date. I don't know why I need to specify that when the topic at hand is a problem I too have experienced arising after the most recent update, but there you have it.
Oftentimes, when I don't have any salient information to share, I just say nothing. "I don't know" is also quite acceptable.
No, here's the thing: everybody tried to help out. I relayed what I found, that I had to click faster than before. You're response of "All you who have NOT (yet) experienced this problem, and therefore felt it would be somehow helpful to respond as if we're all just dimwits who've lost the "skill" of pressing two buttons: It's not. Helpful. Take Apple's advice and Think Different~" was uncalled for and basically insulted everybody who attempted to help you.
The device info is somewhat relevant, since the newer iPhone 7's don't have a home button, didn't know if that was your issue or not. My solution (iPhone 6) was to do press and release quickly, compared to how I did it previously. I had hoped it would be helpful to you, but apparently it was not. I had come to this thread with the same problem you were having. Somewhere along the way, I found that pressing and releasing the home button quicker than before worked as a solution for me.

No, this does NOT work on my iPad Air. Pressing both buttons very fast and at the same time does work, but this is not as it should be - too easy to get the timing, even slightly wrong for it not to work. I use Assisted Touch to bring up the screenshot icon.

Tom, yours has ALWAYS been my method... it only works sporadically now. What has worked on those occasions is pressing *absolutely* simultaneously, or Assistive Touch.
I have noticed that the occasions my typical method (power, home) fails is often when I'm two-layers deep in a game or other app... For example: I'm in a bathroom scene in a game. I click the bathtub, then from that screen, I click the drain. Using my (power, home) method, I can screenshot the game title screen, the bathroom, and the bathtub... but not the drain.
Saying someone's problem is not a problem because you don't have the problem, "It's the same as it ever was" "Nothing has changed" "Just press two buttons, what's so hard", is not helpful. I cannot be any more clear than that. If this does not apply to you, then don't worry about it.
APPLE has officially recognized the problem. It exists. If you have not been affected, consider yourself fortunate.

When I turned Siri off, it worked much easier to hold home and power button simultaneously. A bit of a nuisance, but worked for me.

Its a PITA..... try pressing the home button and the off button simultaneously ....it takes a bit of practice. Unfortunately one of many, many features I dislike in 10.0.2

I found this problem this morning, and this post really helps.
But question is why Apple changing such basic human interface, like my wife couldn't find fast camera access after upgraded to iOS 10.
Apple should keep building interface for consumers NOT for engineers, which Jobs fully understood in the past.

i Agree with you. I am getting fed up with all these so called renewals such most of the time are unnecessary or not helpful. these days I am afraid to install any new update. Problably getting to old for this "****".
i Want a easy working device like the iphone was before.
YOu our really need a week to get around all these new features and changes.
