Does that iPhone happens to be synced with the iTunes Library on that El Capitan volume? Read Sync your iPhone, iPad, or iPod touch with iTunes on your computer using USB - Apple Support. An iPhone can only be synced to one iTunes Library at a time. Perhaps that's the reason Back Up Now is not available?
I'm running iTunes 12.7.1, downloaded today from the Apple website. Before that it was 12.7.1.14 which was part of the High Sierra installation package. Neither version allows backups (the "button" is greyed out). Interestingly, I can run the same downloaded (today) iTunes under El Capitan on another hard drive on my same Mac Pro. This works fine. I moved the backup file produced under El Capitan over to the High Sierra system and iTunes recognizes it. But it won't allow backups!!
None of the sync options are checked on either the High Sierra or El Capitan copies of iTunes. Hard to believe its a sync issue since all worked properly under Sierra, before High Sierra was installed.
Is the latest version of iTunes installed and running on that Mac Pro?
