Here's the commands needed to add multiple dock separators (and yes, it can be used multiple times to create several spacers):

defaults write com.apple.dock persistent-apps -array-add '{tile-data={}; tile-type="spacer-tile";}'

killall Dock

To add more spacers, simply repeat these commands in order.
