When using the Apple TV 4 as a homekit hub , can it be connected true ethernet of has it to be wifi ?
Some tell me wifi others tell me there is no difference
And can a second Apple TV be used as an extender in a big house when on the same network and Apple ID ?

unless ones router is not bridging ethernet and wifi network then it's 100% transparent for all connected devices if they are communicating with a device connected by wire or wireless.
