There probably are applications out there that can do what you want, but I don't know of any off hand. One way you could do it is to import your movie into iMovie and then use the Preview app on your Mac to create transparencies of your arrows and callouts that you can add to your movie using the Picture in Picture feature of iMovie.
Quicktime Pro is not available any more.
I have not used Final Cut so cannot comment on whether it can do what you describe.
To use the Preview app to create transparencies, choose any random photo and make a duplicate of it to work with. Open the duplicate in Preview do an Edit/Select All, Edit/Delete. Select "Convert to PNG" in the drop down window that appears. That will leave you with a blank gray transparancy screen like the one below. Then click on Tools/Annotate/Arrow and you will get an arrow that you can rotate and change the color, thickness, and position of. You also can click on text and type in text.

Save the transparency that you just created. Drag it into iMovie and place it above the point that you want in the clip.
Select the inserted clip. In the tool bar at the top right, click on the overlapping squares icon and choose Picture in Picture from the drop down menu. Adjust the size and position of the insert by dragging the corners of the adjustment rectangle. You can drag completely off the display screen if you want. Then click on the little blue reset button to the right of your screen. That will place the arrow into your movie. You can do this with other transparencies that you create, including inserting text.
