I would like to be able to change my "From" address on a per email basis. I have added an email address amazon@mydomain.com via preferences which I then can select from the "From" drop down list when I compose an email. However, although my Sent folder shows the email having been sent from amazon@mydomain.com it arrives at a 3rd party as myname@gmail.com which is my usual email address.

OK - so how did you add the email address? If you added it as an Alias of your gmail account then that's the behaviour I would expect.
You need to add your other address as a new account. Then it'll set up the proper SMTP server settings

In fact, when I add an email address to an account that has a different domain, it doesn't let me send it at all.
That won't work, because the OP is already choosing the "from" address per email. It appears that the "From" address he's entered is using the Gmail SMTP server, which would indicate to me he's added it as an alias of the Gmail account, not as a separate email account.
Not really - by adding the email address as an alias of your Gmail account, it's using the Gmail SMTP server, which is why it's showing up as Gmail to your recipients.
By adding it as a new account, it uses the Mydomainname SMTP server and your recipients will see the email address as you intend them to...

You can add as many email address to this category as you wish separated by comma.
You do not need to create a new account, just to have multiple address available to you in the dropdown of replying or composing.
In composing a new mail you can choose from the drop down the email address you want to populate the From field.

That's not what the OP is saying. You DO need to create a new account in order for the email to be seen by the recipients as coming from that domain. All you are doing in your "solution" is creating aliases of the one account. They still use the SMTP server settings of the main account
When I came to actually create an account I realised a problem. The email address I want to set as a From address is provided by my domain provider (fasthosts.co.uk) but doesn't actually have an email account associated with it.
In Mail's Accounts preference pane move the my domain email account to the top of the list. Close mail, reboot. It should then be at the top of the From list.
