On my Late 2013 Mac Book Pro (Retina, 15 Inch), 2.6GHz Intel Core i7, 16GB RAM, 1TB SSD, macOS Sierra 10.12.6 previewing an audio file in Finder with Quick Look (hitting the space bar) takes around 15-20 seconds until the playback begins.
Anyone has an idea why this is?
You have so much 3rd party software running, there is no way to tell what might be causing the problem. Try quitting it all and see if that helps.
Try uninstalling CleanMyMac using the Developer's uninstall instructions.
I would uninstall Sophos. It tends to interfere with the computer's operation while providing minimal to no benefit unless you work with Windows files. Most long time posters recommend not using antivirus software or cleaning software such as CleanMyMac.

You really REALLY need to go through the long list of kernel extensions, the long list of Launch Agents, the long list of Launch Daemons and the long list of Login Items. Each and everyone of them is eating up your RAM before you even launch an App you want to use. They will leach your CPU performance as well, not to mention battery life.
Since Safe mode did not really speed up your Quick Look all that much, I would unplug all the external disk, including the SD card you have plugged into your Mac and see what that does for Quick Look.

I wholeheartedly agree with Eric.
Disable all of the items in the User Login Items: and add them back one at a time. Check Quicklook at each stage of the process. Also I'd remove those items that can be launched manually when you really need them. They are just dragging down the system performance all running in the background like that.

The default is on 12K and 10K respectively

Try removing the sysctl.conf (you can rename it so you don't lose it), and reboot. Does that change anything?
If it doesn't change anything, you can always put it back. Or leave it out and see if you really need that many open files per process. Maybe the app that wanted to open 2,000,000 concurrent files is no longer installed, or it has been improved to not need that many concurrent open files.
You are welcome. It may not help, but try running posting the Etrecheck report.
By default, no /etc/sysctl.conf exists. You have one. Have you, or any of the installed applications decided to modify kernel behavior, or max/min settings in this file? Aside from what everyone else has observed, changes in the /etc/sysctl.conf file can rear up and effect system performance too.
Thanks for your quick reply! I tried the Safe Mode, the quick look "reacts" a bit faster, but it still took quite a long time from pressing the space bar until playback started... I'd say it takes around 5-10 seconds in Safe Mode
Thank you Bob... I'll do some "housekeeping"... Already removed Sophos, but this didn't change the behaviour. I'll do some more and will let you all know, once the "problem" is solved. I have read somewhere that resetting the SMC helped, but not in my case.
