Do photos and videos count against my storage space in iCloud?

If you turn on Settings > [username] > iCloud > Photos > iCloud Photos they will sync to iCloud (and to any other device where that option is turned on) and they do count against your iCloud storage.

If you do not turn on iCloud Photos but do turn on Settings > [username] > iCloud > iCloud Backup, they will backup to iCloud and they do count against your iCloud storage.
If you don't turn on either of the above but do turn on My Photo Stream, they do not count against your iCloud Storage. My Photo Stream is an older service that is no longer to some newer Apple IDs and is designed primarily for syncing photos to other devices. It only keeps the photos for 30 days. I don't quite understand.
What is a photostream and how is that different from the photos I take on my iphone and stored on my iphone?
If I take photos and videos on my iphone and they are backed up to my iCloud account, do those count against my storage?
Photos in iCloud Photos and in iCloud Backup do.
