To upgrade from Catalina to Big Sur should I remove Microsoft office and my printer application?

I didn't and I had no issues. Also I have not seen any recommendation from Apple to do that. Upgrade instructions are in How to upgrade to macOS Big Sur - Apple Support

Neither is required. But if you need more space for the installation it might be necessary.
