Conserving battery health

To keep my MacBook battery healthy, is it better to plug in my MacBook whilst I'm working, or to let it run off the battery?

With modern technology the battery can self detect whenever its full and if it is full and the charger remains plugged then it will bypass the battery and feed your laptop in short terms the laptop stops using the battery but uses the charger instead its around the same as using regularly but I wouldn't recommend using it dry to 0%. But if you want to keep it healthy try to shut it off when your not using, Avoid extremes of heat and cold. You should read this too https://discussions.apple.com/thread/3818265.
