I go into Mail, Preferences, Fonts & Colours. Press select and nothing happens. I see this is an issue that has happened to others. I have quit Mail and come out again. I have looked in Library for .com.apple.mail * and there is nothing in the .com area of library with anything to do with Mail.
What next?

Where are you wanting Font size to increase?
In individual incoming mail this is can easily be done using the Mail>View>Customize Tool Bar and add the Smaller/Bigger option. You can repeatably hit Bigger...
