External drive not working after trying to use it for Time Machine – Help!

I have a fairly new Lacie Rugged 5TB drive that was working perfectly fine and i wanted to set it up as my Time Machine drive. I selected the drive as my backup disc in Time Machine but immediately got the following error message:

Now the Lacie drive doesn't even mount on my desktop! It appears in disc utility as 'disk2S2' but again, doesn't mount or appear on my desktop.

I tried using the mount function in disk utility but got this error message:

I tried running disc first aid and got this error message:

Any ideas what's going on here or how to get my drive back to life? Like i say the Lacie drive was almost new and not cheap!!

My setup:

Thanks
