I figured it out.

It was Zoom

The app was still open in the dashboard and it appears you don't have to be actively using it or in the window for it to override your other shortcuts.

The normal function of Command+T from the desktop is: Open a new tab in a web browser and other applications that support tabs. So it's not the OS that's interfering.

I've been using Command+T in Photoshop without issue in Big Sur. Does what it has always done - calls up a transform.

I notice the box is also telling you what size of an area it's going to capture, 2560*1415. This isn't anything the OS does, including the newer Screenshot utility. I would have to assume you have some sort of third party app installed that's getting in the way.

Edit: Ah. You figured it out while I was typing.
