Mail signature problem after upgrading macOS Big Sur 11.4

Recently I upgraded my MacBook Air to macOS Big Sur 11.4 and now I could see issue of my mail signature is not working.

Create an email signature in Mail on iCloud.com

You can create a plain text signature that appears automatically on every message you send.

Saying Signatures does not work is too vague. Can you give more details and actions taken ?
