Yes this option will not be available unless ‚ Dark is preselected in the >System Preferences>General
Then you can make changes in Mai.app>Preferences> uncheck the box obviously

Use a light or dark appearance on your Mac - Apple Support
Does the computer have Dark Mode as the Default Setting. Check in System Preferences >> Desktop & Screen Saver >> Desktop tab and change to Light Mode or Dynamic Mode instead of Dark Mode. - Then test.
Am working in Catalina right now - and do not see anywhere is Apple Mail Preferences an option to make the Preview Panel While vs Black or any colour at all.

Mail Preferences > Viewing > Uncheck use dark backgrounds for messages. This will produce white message backgrounds even when the Appearance mode is set to Dark in System Preferences > General.
Your answer was very helpful but I specifically want to change the background on ‘mail’ from black to white.
