Can a backup from 12.5.4 be used to restore on new iPad Pro?

If I backup and encrypt a backup through iTunes, of an iPad only able to install 12.5.4, will I be able to “restore from backup” on a 2020 iPad Pro that I purchase. Assuming the new ones come with iOS 14 pre installed.

In short, yes you can.

In greater detail… an iCloud or iTunes backup can be restored to a device that has the same, or later, version of iOS/iPadOS as that which created the backup.

Put another way, you cannot restore a backup to a device that has an earlier version than that of the iCloud/iTunes backup.
