Indeed, if set up and you select Speak Screen from the Share menu Siri reports, "No speakable content could be found on the screen". However, you can use an onscreen controller to access Speak Screen. You’ll need to use the Kindle app for iPad; this won’t work with Kindle Cloud Reader in a web browser.
Navigate to Settings > Accessibility > Spoken Content. Toggle on Speak Screen. Tap Voices if you wish to select a different speaking voice. Otherwise, tap Speech Controller directly below, then toggle on Show Controller. If desired set Idle Opacity. The range is 5-100%. Tap Double Tap below, then tap Read All Content. Tap Speech Controller to return to the previous screen. Tap Spoken Content to return to the previous screen. You can leave the Settings app open for easy access to toggle off Speak Screen.
Open the Kindle app. Open a book. The Speech Controller floats and you can move it anywhere to an edge of the screen with a press-drag action. Double-tap the controller to initiate Speak Screen. Single-tap the controller to expand. Here you can control play/stop, go to the previous or next page and set the speaking rate. The default range is 1/2x to 2x. You can ignore the speak selection control as it doesn’t function as designed in the Kindle app. Tap the circular "<" button to collapse the controller. The controller will timeout and collapse automatically after approx. 8s. To dismiss the Speech Controller, return to the Settings app and toggle off Speak Screen.

Hey Smcg1379,
Thanks for posting your question in Apple Support Communities.
The format of the Kindle books may not meet the requirements needed for the word to be read by the iPad. You can try checking for any updates to your iPad and the Kindle app. Ultimately, you may need to reach out to Amazon Kindle support to find any options available for the audiobook function.
You can find ways to update your apps in this article: How to manually update apps on your Apple device.
If you need to contact Amazon for support, you can find way to contact app developers in this article: How to contact an app developer.
Best Wishes.
