How can this pop up on my screen if pop up blocker is enabled? Clearly I shouldn’t click on it? How do I check it my phone is infected?

It is a scam notification. Your iPhone is not infected with anything. The perpetrator wants the user to click on the supplied link which will lead into the App Store so that the user can buy a useless app. Ignore that notification and close the page.
