You would not be able to transfer all of your photos to iCloud and not have them on your iPhone. ICloud Photos instead syncs your photos to iCloud and all devices you are signed into. Please see: Set up and use iCloud Photos.
If you delete your photos from your device, they will be deleted everywhere when you use iCloud Photos.
If you are running out of storage on your iPhone, select Optimize iPhone Storage in Settings > Photos.
If you aren't using iCloud Photos, then they would be included in your backup: What does iCloud back up?
