If we understand correctly you're unable to reply to incoming calls using Messages on the Lock Screen.

Does this seem to occur over a stronger cellular signal?

Can you send messages as expected when you're not using Phone for calls?

Do you have the most recent version of iOS 15.1 installed? You can also check and make sure you're carrier details are updated as well.

Should the issue continue you can reach out to Apple Support directly.
