If you have an issue when you are logged in and trying to use keyboard short cuts/trackpad gestures for Mission Control—To trouble shoot further you can:
—Try a SafeBoot https://support.apple.com/en-us/HT201262
Takes noticeable longer to get to the login screen, does a 5 minute disk repair before it fully boots up, and certain system caches get cleared and rebuilt, including dynamic loader cache, etc.
Login and test. Reboot as normal and test. Caches get rebuilt automatically.
In Safe mode third party system modifications and system accelerations are disabled, it removes malware, etc hampering smooth operation, however a reboot will put it back to normal mode.
This test will tell you if third party interference; extensions etc are not loaded in safe boot mode.

This will tell you if it a universal issue or isolated to your user/admin account.
Uninstall all third party apps that are Cleaners/Optimizers/VPN/Anti-Virus
all known to cause issues on the macOS

I have the same problems: Sometimes mission control no longer works after the MacBook wakes up from sleep. I have a MacBook Pro 16 with M1 chip. Is there any possibility to re-initiate mission control? Is there a process I can kill and then start again to make mission control work again?
I've been able to get it to work again by going to Displays, changing the resolution, and then changing it back. Very annoying bug though and it only happens on one of two external monitors I've got plugged in.
Duh, of course, it doesn't work in sleep mode! It's when I wake it up that it stops!
Start up in Safe Mode as in the above description and it works flawlessly
