Can I backup from two different drives (one external, one internal) to my Time Machine drive?

Open the Time Machine pane of System Preferences, click on Options, and remove the external drive from the exclusion list. If it's grayed out, not without reformatting it as APFS or Mac OS Extended (Journaled).
