Screenshot is not letting me choose clipboard as the save location
I used to be able to select clipboard as the save location but it is not showing, why? How do I fix it?

FWIW, I just had a closer look at your screen shot. You have "Record Entire Screen" selected. You can't copy a video recording to the Clipboard. That's why it's not in the list.

You should still see Clipboard as an option…
Try booting into Safe Mode, then restart normally.

Where to find screenshots
By default, screenshots save to your desktop with the name ”Screen Shot [date] at [time].png.”
In macOS Mojave or later, you can change the default location of saved screenshots from the Options menu in the Screenshot app. You can also drag the thumbnail to a folder or document.
